Biopolymer/Genomics: DNA Sequencing

Biopolymer/Genomics
Shared Service

Applied Biosystems Model 3730XL high throughput 96-capillary and Model 3100 16-capillary DNA sequencers support the Biopolymer/Genomics Core Facility’s DNA sequencing service.

We offer full-service DNA sequencing. Investigators need only provide the DNA templates and any custom primers they want used in the sequencing reaction. We will perform and run the sequencing reaction and provide the data electronically and as a printed copy. Alternatively, investigators may perform their own sequencing reactions, perform post-reaction clean-up, and submit as "ready-to-run" samples at a significantly reduced rate.

As an added service to those investigators who prefer to run their own reactions, aliquots of Applied Biosystems’ Big-Dye terminator V3.1 reaction mixture can be purchased at our reduced bulk rate. We have an extremely large collection of primers specific to a variety of vectors. Please contact us to find out if we have primers for a specific vector you have before submitting a request to have them synthesized. (A partial listing can be found at the following link ____) The facility has extensive experience sequencing DNA from a variety of sources including plasmids, PCR products, lambda or other bacterial virus vectors, BACs and even genomic DNA.

We also offer primer walking of large DNA sequences, although some investigators prefer to analyze sequence data and design their own primers to continue sequencing. The facility utilizes the software package Sequencher to analyze and construct large sequencing projects. Many investigators who routinely sequence large DNA fragments or have need for multiple alignments have purchased their own personal copies. This facilitates data transfer from the facility to the investigator. We can accommodate any of the dye-labeling sequencing kits that Applied Biosystems sells, or the ET dye kits sold by Amersham (now GE Healthcare).

The cost for DNA sequencing is:

Full Service: (Core does the sequencing reaction, clean-up, run the reaction and provide data)
$12.00 per reaction for single reactions
$1000 per 96 well tray

Ready To Run Samples:
$5.00 per sample (Maximum of $140.00 if all submitted at once)
$140.00 per 96 well tray

Cost for Primer design and synthesis:
Current price per base for synthesis at 40nMole without purification (currently $0.40 per base).
